I use a simple mouse as my main tool, and Sai for sketching (if not done by hand), linearting, coloring and everything else, and use PSD (CS2) exclusively for some final touches, Speech texts, and the signature if required. Can't really suggest any program since these are the only ones that work fine for me since I'm not a graphic tablet user.

I use CSP ex/ manga studio ex 5. It does really well for all my artwork. It also makes it easy to do doujin, manga and comics. Also simple animation too. The program is a little complex at first and even 2 years of drawing I'm still learning new things and easier ways to do things. I also use a Wacom intuos pro medium size.
